Antibodies in VR: New Alzheimers disease drug, aducanumab (Adulhem), bound to beta amyloid plaque

In this video, we discuss a new drug from a biotech company called Biogen that was just recently approved by the FDA for treating Alzheimer’s Disease. It’s the first drug cleared by the FDA to slow cognitive decline in people living with Alzheimer’s and the first new medicine for the disease in nearly two decades. The FDA’s decision was controversial but in our video we’re only going to discuss the mechanism of action of this drug, which goes by the name aducanumab. Aducanumab targets a “sticky” compound in the brain known as beta-amyloid, which most scientists expect plays a role in the devastating disease. Biochemical and structural analyses show that aducanumab binds a linear epitope formed by amino acids 3–7 of the beta amyloid peptide. The crystal structure of the Fab fragment of aducanumab bound to its epitope peptide reveals that aducanumab binds to the N terminus of beta amyloid in an extended conformation, distinct from those seen in structures with other antibodies that target this epitope.
